Metabarcoding in the Thau and Berre lagoons to detect Veneroidaee
In the framework of the PALDIAG project, environmental DNA from seawater was gathered in 2023 in Thau and Berre lagoons and studied with a 16S marker in order to detect the Veneroidae species.

Seawater was filtered in June-July 2023 in Thau and Berre lagoons. The filtering capsules (24) are Waterra systems using a customized pumping system commercialized by Argaly company. DNA was resuspended in the capsule vigorous agitation after the injection of Longmire buffer, and extracted at Argaly. Whole DNA was used to construct libraries of metabarcoding using 16S primers specific to Veneroidae. Sequencing was performed on a MiSeq sequencer (2X 150 bp). Besides an experiment was conducted in October 2022 in Ifremer Sète and 14 additionnal capsules were obtained from the filtering of water in aquariums filled with different ratio of two clam species. |
